The a2b1 integrin serves as an extracellular matrix receptor for collagen and /or laminin, as well as several other nonmatrix ligands.1-3 Interactions between the a2b1 integrin and the extracellular matrix have been implicated in a number of biologic and pathobiologic processes.4 The important role that the a2b1 integrin serves as a collagen receptor on platelets for platelet function and hemostasis is well established.4-7 In some cell types such as endothelial cells and epithelial cells, a more activated form of the receptor may serve as both a collagen and laminin receptor.3,8 Extensive in vitro studies on pulmonary, gastrointestinal and breast epithelia suggest that signals downstream of a2b1 integrin ligation contribute to branching morphogenesis.9-12 Experiments using explant cultures and 3-dimensional collagen gels have implicated the a2b1 integrin in keratinocyte migration and fibroblast contraction during wound healing.13-15 The role of the a2a1 integrin in these multiple complex biologic processes will be discussed in this chapter. system. collagen subtypes.
