IL-10 is a pleiotropic cytokine that is produced by a variety of hematopoietic cells (e.g., CD4+ T cells, NK cells, macrophages, dendritic cells)1-4 and regulates the function of several adaptive immunity-related cells. Although generally considered an immunosuppressive molecule, IL-10 has been demonstrated to possess immunostimulating properties in several in vitro and in vivo models. These very different outcomes are believed to depend upon experimental conditions (e.g., in vitro/in vivo), dominant effector system involved in a given immune response (humoral immunity, Th1 type immunity, mixed innate/adaptive immunity), timing of IL-10 production/ administration, and IL-10 dose and/or location of expression. However, the cascade of molecular events underlying IL-10 ultimate effect on adaptive immune responses are still largely unexplored.
