Annexin structures have been found to show a common fold due to their characteristic topology. The structural conservation within the annexin family throughout the animal, fungal and plant kingdoms is described based on X-ray crystallographic structures of wild type annexins and their complexes. Structural aspects of membrane-bound annexins are discussed on the grounds of electron microscopical data as well as an extensive biophysical literature. To date, it seems that the membrane-bound form of annexins is also the interface to their physiological functions. This is demonstrated by some recent findings regarding annexin biology in extracellular locations and the phenomenon of amino-terminal regulation. We finally conclude that annexins present allostery as a common feature and possess a high potential of allosterical regulation.
